Historical Development:
The inception of SR9009 can be traced back to the early 2010s, when it was developed by the research team at The Scripps Research Institute. SR9009 was initially synthesized as part of a series of compounds aimed at investigating the modulation of circadian rhythms. The compound targets Rev-Erbα, a nuclear receptor involved in regulating the body’s internal clock, lipid and glucose metabolism, as well as inflammatory responses.
Chemical Structure:
Chemically, SR9009 has a molecular structure that comprises a pyrrolidine backbone with an ethyl carboxylate group at one end and a complex chlorobenzyl-nitrothiophene-methylamine moiety attached at the other end. This chemical structure is unique and not steroidal or related to conventional small-molecule pharmaceuticals.
Properties and Mechanism of Action:
SR9009 is a synthetic Rev-Erbα agonist. By binding to and activating Rev-Erbα, SR9009 influences various biological processes regulated by this receptor. Rev-Erbα plays a significant role in the circadian regulation of metabolism, and its activation leads to alterations in the expression of genes involved in lipid and glucose metabolism.
Notably, in preclinical studies, SR9009 has shown to enhance metabolic activity in skeletal muscle, resulting in increased endurance and reduced adiposity. The compound also exerts anti-inflammatory effects and modulates mitochondrial content and function.
