The Origins of YK-11
YK-11 appeared in research circles earlier this decade, raising attention with its unique profile. In the early 2010s, Japanese scientist Dr. Yuichiro Kanno was researching compounds that might build muscle in a safer way than traditional steroids. During testing, he realized YK-11 was special. It powered up muscle growth not by just mimicking testosterone like other SARMs, but with a sneaky two-step process:
Step 1) It blocks myostatin, a protein that typically puts the brakes on muscle growth.
Step 2) It boosts levels of follistatin, a different protein that stomps on myostatin and kicks muscle growth into overdrive.

How YK-11 Works Its Magic
When you look at the chemical structure of YK-11, you’ll notice it looks a lot like traditional androgenic hormones like testosterone or DHT. But peek closer—it has some special alterations that make it unique.
Namely, YK-11 contains both:
● A trifluoromethyl group
● A carboxylic ester
These molecular tweaks give YK-11 exceptional “binding affinity”—in plain English, it sticks like glue to androgen receptors in muscle and bone. And once it locks on, it sends signals directing those cells to grow bigger and stronger.
This selective and targeted action is what separates YK-11 from normal SARMs and makes it such an intriguing product. Early research indicates it builds muscle and strength without excess side effects, but more studies are still needed to verify these results.
YK-11 Research Studies
Although YK-11 never passed the pre-clinical development stage – there are a couple of early studies that show it’s potential for muscle and bone enhancement, along with increased strength and performance.
One of those studies, published in 2013, investigated how YK-11 induces muscle cell differentiation. Researchers treated C2C12 muscle cells with YK-11 or the androgen dihydrotestosterone (DHT). They found YK-11 more potently caused the expression of key muscle differentiation genes like MyoD, Myf5, and myogenin compared to DHT. Interestingly, YK-11, but not DHT, increased muscle growth-promoting protein follistatin levels. Using an anti-follistatin antibody, the researchers showed follistatin induction is essential for YK-11’s muscle-building effects. This also proved that YK-11 is a potent myostatin inhibitor.
A second study was published in 2018 that looked at the effects of YK-11 on bone cell development. Researchers treated osteoblast bone cells with YK-11 or the androgen dihydrotestosterone (DHT). They found both YK-11 and DHT accelerated bone cell proliferation and mineralization. However, YK-11 treatment more strongly increased levels of proteins linked to osteoblast differentiation like osteoprotegerin and osteocalcin compared to control untreated cells. These effects were blocked by an androgen receptor antagonist, indicating they depend on the androgen receptor. To understand how YK-11 works, the researchers looked at rapid, non-genomic androgen receptor signaling. They found YK-11, similarly to DHT, activates a protein called Akt linked to osteoblast differentiation. So they concluded YK-11 has osteogenic activity mediated in part via androgen receptor activation of Akt-signaling. This highlights a new mechanism for promoting bone cell development by this intriguing research SARM.
